can one device be VMDirectPath twice?

 can one device be VMDirectPath twice?
This is probably a dumb question as I know little on VMDirectPath.
Say I have a RAID card and want to try different NAS software, can I create two VM, and have both passthrough to the same RAID card?

No, unforturately a device can only be used by a single VM at one time.
You could configure both VMs to use the device, but only one could be powered on at a time.

One more question:
Once the device is passthrough to a VM, can it be used/accessed indirectly via hypervisor by another VM or even by hypervisor itself?

Once you power on the VM, the device can't be used by the hypervisor or another VM.
Once it is powered off you can use it with another VM. To use it with the hypervisor you would need to disable the device for use with VMDirectpath and reboot.
